In Paul Buentello debut in the UFC, on February 2, 2005, he had won over Justin Eilers with a KO in 3:34 of the first round for his debut fight in UFC 51: Super Saturday. Paul won the next fight with Kevin Jordan by guillotine choke submission on June 4, 2005, on UFC 53: Heavy Hitters at 4:00 of the first round.

On October 7, 2005, Paul lost a title shot against  Andrei "The Pitbull" Arlovskiwho is a heavily-touted Interim UFC heavyweight champion at UFC 55 and Arlovski defeated him with a 15-second knock-out. Following his loss to Arlovski, he fights against Gilbert Aldana at UFC 57 where he won through TKO due to strikes.

After leaving the UFC, Paul fought for Strikeforce in which he won 3 matches straight against lesser competition, before losing to Alistair Overeem in a bout for the Strikeforce heavyweight champion.

Then, Paul moved to a new major organization, Affliction, and he won his first two bouts. At the Texas-based Shark Fights promotion, Paul Buentello began again his Mixed Martial Arts career, defeating the previously unbeaten (6-0) Bryan Humes through Unanimous Decision. On August 14, he fought against Tim Sylvia, a former UFC Heavyweight Champion at Powerhouse World Promotions: War on the Mainland and he lost the fight through KO in the second round.

On November 13, 2010, Paul Buentello was scheduled to face Kerry Schall at Nemesis Fighting: MMA Global Invasion but the event was postponed to avoid a tropical storm on December 10, 2010.
