Early in her career, Vaughn acted in movies like Friday and Don't Be a Menace to South Central While Drinking Your Juice in the Hood. She played alongside Joan Severance in the comedy-action film Black Scorpion (1995) and its sequel, Black Scorpion II: Aftershock (1997). In 1997, Vaughn got a role as Lovita Alizay Jenkins-Robinson, the high school secretary, in the second season of The Steve Harvey Show on The WB. She received three NAACP Image Awards for Outstanding Supporting Actress in a Comedy Series for her work on the show, which ended in 2002.

Later in 2002, Vaughn joined the cast of the Showtime drama series Soul Food as Eva Holly and received another NAACP Image Award nomination for Outstanding Supporting Actress in a Drama Series. From 2003 to 2005, she was a regular cast member on the UPN/The CW sitcom All of Us. In 2007, she played a supporting role in Tyler Perry's Daddy's Little Girls, and later, she had a regular role in another of Perry's productions, Meet the Browns (2009-2011).

Vaughn is also the owner of a production company called Nina Holiday Entertainment, which produced films like Sugar Mommas, A Cross to Bear, and Girlfriends' Getaway. In 2016, she made her directing debut with the comedy film #DigitalLivesMatter. In the same year, Vaughn took on a recurring role as a housekeeper in the Oprah Winfrey Network drama series Greenleaf, alongside Keith David and Lynn Whitfield. In 2020, she had a recurring role as Robin in the South African TV series Kings Of Jo'Burg, which is shown on Netflix.
